Das Ziel dieser Dissertation war die in vitro Rekonstruktion verschiedener vaskulärer Barrieren. Als in vitro Modell für ein vaskularisiertes, dreidimensionales (3D)?Gewebe wurde der mikrofluidische ?3DVasc Bioreaktor eingesetzt. Dieser auf der SMART?Technologie basierende Bioreaktor besteht aus einer porösen Mikrokapillare zur Kultivierung von Gefäßendothel und einer darunter liegenden, die Kapillare umgebenden, Kammer für ein 3D?Gewebe. Drei Barrieren wurden ausgesucht: die Blut?Hirn?Schranke, das Darmepithel und die Leber. Alle Modelle wurden 3D etabliert und im Vergleich zu den 2D?Modellen und den natürlichen in vivo Barrieren validiert.Die in dieser Arbeit vorgestellten 3D?Modelle im ?3DVasc Bioreaktor bieten in Zukunft eine geeignete Plattform zur Untersuchung der Wirkstoffaufnahme sowie der Verteilung und der Anreicherung der applizierten Substanzen in Geweben. Die Verknüpfung der drei Modelle untereinander würde zahlreiche innovative Untersuchungsmöglichkeiten schaffen, wie z.B. neu entwickelte Substanzen effizient und kostengünstig in einem HTS Format zu testen, um geeignete Kandidaten schneller zu bestimmen, weiter zu optimieren und letztendlich auf den Markt zu bringen.
