This book describes characterization of the intestinal absorption and bioavailability of ?-tocotrienol using in vitro, in vivo and in situ models..... investigates the carrier-mediated transport of ?-tocotrienol by NPC1L1 and compares its oral bioavailability, oral absorption and elimination kinetics to that of ?-tocopherol....characterizes ?-tocotrienol and ?-tocopherol in vitro interaction kinetics with NPC1L1 transporter responsiple for both compounds non-linear kinetics....tests solid lipid nanoparticles as a model formulation to enhance the permeability of ?-tocotrienol and investigates the mechanism of intestinal uptake of ?-tocotrienol packaged in this delivery ssytem..... conducts preclinical pharmacokinetic tests required to predict the oral absorption kinetics of UK-343, 664 (a substrate for P-gp and CYP3A4) in humans....and builds an in silico PBPK simulation model to investigate and explain the non-linear absorption kinetics of UK-343, 664 obsrved in humans using the ACAT model (Gastroplus™, Simulation Plus Inc., Lancaster, CA), and compare the simulation results with the in vivo results.
